Photopolymerized Multifunctional Hydrogel Loaded with Asiaticoside and Growth Factor for Accelerating Diabetic Wound Healing

Diabetic wounds are difficult to heal for a long time because of their complex bacterial infection, excessive inflammation, and angiogenesis obstruction. An effective treatment strategy is desirable to accelerate diabetic wound healing. Herein, a highly adhesive, stretchable, self-healing, and antibacterial multifunctional hydrogel loaded with asiaticoside (AS) and recombinant basic human fibroblast growth factor 2 (FGF-2) in situ is rapidly prepared by photopolymerization under green light-emitting diode (LED) irradiation within 80 s. The hydrogel exhibits high stretchability, outstanding tissue adhesive properties, and good self-healing properties. Moreover, the hydrogel exhibits excellent biocompatibility, enhanced cell proliferation and migration ability, and broad-spectrum antibacterial activity. The hydrogel can effectively modulate the continuous release of AS and FGF-2 to enhance the antibacterial, anti-inflammatory, and angiogenesis effects to synergistically accelerate diabetic wound healing. Notably, the hydrogel can effectively enhance the ability to promote angiogenesis and collagen deposition and significantly shorten the healing time of diabetic wound in rats. Therefore, it is an effective strategy to prepare a multifunctional hydrogel loaded with AS and FGF-2 by photopolymerization under green LED irradiation for accelerating diabetic wound healing.
