Abstract DNA-protein crosslinks (DPC) induced by endogenous aldehydes are highly toxic DNA lesions that interfere with replication and transcription. To investigate how cells overcome DPC lesions and to understand the cellular toxicity of aldehydes and their impact as DPC in living organisms, we established a high-throughput sequencing method to explore the genome-wide distribution of DPC (DPC-seq). This protocol enables the measurement of DPC distribution and repair kinetics using cells obtained from living animal organs or tissue culture samples treated with formaldehyde.
