A benzothiazole derivative for differentiating mitochondrial G-quadruplex formation between normal and tumor cells

Emerging studies demonstrates that mitochondrial G-quadruplex (G4) may be a prospective diagnostic and therapeutic target for cancer. Fluorescent probes targeting mitochondrial G4 are crucial for in-depth research on G4 function and mechanism and developing new diagnostic techniques. Here, we report a benzothiazole derivative named ThT-E, which can be prepared with high yield through only one step of reaction. ThT-E exhibits superior detection sensitivity compared to existing probes, and also demonstrates reliable performance for in situ characterization of mitochondrial G4 in living cells. Interestingly, ThT-E has been used to differentiate tumor cells from normal cells by quantifying the formation of mitochondrial G4s, implicating that ThT-E may have wide applications in tumor diagnosis.
