A Flow Cytometry Evaluation of Poly (1-vinyl-3-alkyl imidazolium) Based Ionic Liquid's In Vitro Effect on Various Cellular Components of Blood

This study aims to gain insight into the effect of the synthesized poly-ionic liquids (PILs) (1-vinyl-3-alkyl imidazolium bromide) (alkyl=hexyl (A), octyl (B), and decyl (C)) on various cellular components of blood by flow cytometry. PILs may have some adverse effect on blood depending on the nature and the dose administered. So, it is critical to understand the potential influences of foreign polymeric entities on blood before they are used clinically. The complete blood counts (CBCs) are done using an automated analyzer based on the flow cytometry principle. The leukocyte viability with different PIL samples is performed by flow cytometry using 7-AAD. The auto-hemolysis is performed by incubating the PILs with defibrinated blood. The CBC is mostly the same when incubated with various PILs. However, RBCs show an increase in mean corpuscular volume (MCV), indicating cellular swelling. The viability with phosphate saline buffer (PBS), dimethyl sulphoxide (DMSO), and PIL (A, B, and C) is 99.94 %, 99.76 %, 99.71 %, 98.87 %, and 99.42 %, respectively. The effect on leukocyte size & internal structures are noted. The auto-hemolysis tests of RBC by PILs are very similar to control auto-hemolysis. This study aims to gain insight into the effect of the synthesized poly-ionic liquids (PILs) (1-vinyl-3-alkyl imidazolium bromide) (alkyl=hexyl (A), octyl (B), and decyl (C)) on various cellular components of blood using flow cytometry. The in vitro analysis indicates that PILs neither alter the cell counts of blood nor the viability or auto-hemolysis to a significant extent.image
