•A P. aeruginosa RPA-CRISPR/Cas12a detection platform using lasB gene was established.•The platform is well contained and exclusive.•The one-tube RPA-CRISPR/Cas12a assay effectively reduces aerosol contamination.•Portable fluorescence detectors, UV, and blue light irradiation yield results.
