Diagnostic Probes Designed for Detection of Mycobacterium Tuberculosis - An Insilico Approach

An insilico diagnostic probes is designed for the detection of latent Tuberculosis caused by Mycobacterium Tuberculosis. Identification of a specific target is required to diagnose the condition of latent Tuberculosis. Till date, there is no specific detection of latent Tuberculosis. Hence, we have proposed a molecular detection method that will precisely diagnose and detect latent Tuberculosis in a susceptible individual. In this paper, we have designed the diagnostic kit of primers and probes from the genes (embC, pimB, lpqW, dprE1) which are responsible for the expression of ManLAM, a glycolipid present in the region of a capsule of Mycobacterium accountable for its virulence. The designed gene-based oligos and primers will be used to detect latent Tuberculosis via rtPCR and biosensing techniques as they are precise to the genes expressed during ManLAM formation in Myco bacterium Tuberculosis. The oligos and primer pairs are designed and simulated by using PrimerBLAST and Primer 3 Software. Further, the cross-homology with other bacterial species is done using BLAST Nucleotide, confirming that the identified conserved areas do not differ from other bacterial species. The primers and internal oligos have a length of 20 nucleotides each with GC percentage in between 50–60, and self-complementarity is less than 3 nucleotides, which signifies their sensitivity and specificity toward the target. The prognostic/diagnostic kit can be used for latent and active Tuberculosis identification test.
