Aerobic Exercise Delays Serum Metabolite Changes In Neural Aging In The Rat Brain

PURPOSE: To investigate the changes of serum metabolites in brain neural aging in rats delayed by 8 weeks of aerobic exercise. METHODS: A total of 45 8-week-old male SPF-grade SD rats weighing 200 ± 10 g were selected and randomly divided into: normal control (C) group, aging control (A) group and aging exercise (AE) group, 15 rats in each group. The normal control (C) group was treated with saline and the senescent (A) and senescent exercise (AE) groups were treated with intraperitoneal injection of D-gal at a daily dose of 100 mg/kg for 6 weeks . After 1 week of acclimatization, the rats in groups C and A were fed naturally and the rats in group AE were subjected to aerobic interval training (AIT) on a running platform for 5 days per week for 8 weeks. Tail vein blood was collected 1 day before and 8 weeks after exercise and stored at -80 °C for subsequent use. after the exercise intervention in the AE group, Morris water maze experiment was adopted in response to the ability of rats to acquire learning and memory for the water maze and the ability of rats to retain memory for spatial locations. RESULTS: 1) Results of the Morris water maze spatial exploration experiment: group A showed a significant increase in escape latency (p < 0.01) and a decrease in the number of platform crossings (p < 0.01) compared to group C; AE showed a significant decrease in escape latency (p < 0.05) and an increase in the number of platform crossings (p < 0.01) compared to group A.2)Changes in serum metabolites in the tail blood of rats eight weeks after exercise: among them, L-Cystine, Glyceric acid, Pyridoxal, 1,2-Di-(9Z-octadecenoyl)-sn-glycero-3-phosphocholine, Bilirubin, DL-Serine Glycine and other serum metabolites AE group followed the same trend as C group and were all greater than A group. In addition, serum metabolites such as Cysteine-S-sulfate, L-Tryptophan, and cis-Aconitate had the same trend as group C in the AE group, and all were smaller than group A. CONCLUSIONS: 1) 8 weeks of running table exercise improved the spatial learning memory ability of rats .2) 8 weeks of aerobic exercise delayed neural aging of rat brain may be associated with L-Cystine, Glyceric acid, Pyridoxal, 1,2-Di-(9Z-octadecenoyl)-sn-glycero-3-phosphocholine, Bilirubin, DL-Serine, Glycine and decreased Cysteine-S-sulfate, L-Tryptophan, cis-Aconitate.
