Benzoxaboroles have attracted extensive attention in biomedicine because of their unique physicochemical and drug-like properties. Benzoxaboroles were frequently used as essential pharmacophores to target clinically relevant targets and manifest antibacterial, antifungal, antiinflammatory, and anticancer activities, revealing their potential as a privileged scaffold for drug discovery. We summarize benzoxaboroles by focusing on their biological activities and interaction patterns with target proteins. The unique feature of benzoxaboroles, which is that it reacts with a nucleophile from enzyme active site residues or metal-activated water molecules to form a stable tetrahedral intermediate, is highlighted.
