Whole-Genome CRISPR Screening Reveals Genetic Vulnerabilities and Therapeutic Targets in Malignant Pleural Mesothelioma
Journal of Thoracic Oncology(2023)
摘要
Malignant pleural mesothelioma (MPM) is aggressive cancer with a poor prognosis, primarily due to the limited number of effective therapies. Pathologically driven by genetic alterations inactivating pharmacologically intractable tumor suppressor genes (TSGs), MPM has long epitomized cancer recalcitrant to molecule-targeted therapy. Functional genomics enables unbiased interrogation of cancer genomes and is a powerful platform for anticancer drug discovery. However, CRISPR/Cas9-based systematic interrogation of MPM genomes and functional characterization of MPM-specific vulnerabilities remains elusive.
