Despite large-scale genomic analysis of ovarian cancers, common oncogenic drivers have yet to be definitively described. This is largely due to the primary mutational mechanism in ovarian cancer being copy number alteration rather than missense mutation. Although recurrent gains and losses have been identified, they often involve large genomic regions harboring dozens to hundreds of genes, precluding a straightforward candidate validation approach. Our goal was to identify drivers of tumor progression using the Sleeping Beauty (SB) transposon mutagenesis system in a panel of ovarian cancers of varying subtypes in an in vivo xenograft mouse model.
