Abstract
Topic: 3. Acute myeloid leukemia - Biology & Translational Research Background: Acute leukemia(AL) is a modern disease, but there are records about this kind of diseases in the classical Chinese medicine books thousands of years ago. The description of yin and yang toxicosis in the classic Chinese medicine book “Jingui Yaolue” conforms to the clinical manifestations of acute leukemia. Its prescription for treating yin and yang toxicosis is Shengma Biejia Decoction. The formula contains realgar(As4S4), an important drug for the treatment of acute promyelocytic leukemia. In China, Shengma Biejia Decoction has been frequently used in the treatment of acute leukemia and has achieved excellent clinical efficacy. However, its mechanism of action is not fully clear. Aims: To investigate the regulatory effect of Shengma Biejia Decoction on mitochondrial mitosis-associated proteins Drp-1, Fis-1 and mitochondrial autophagy-related proteins Parkin and PINK1 in human acute leukemia (AL). To explore the effective mitochondrial dynamics mechanism of the traditional Chinese medicine for acute leukemia. Methods: The human acute myeloid leukemia model was established with KG-1a cells on the NSG mouse platform. The mice fed with Shengma Biejia Decoction were the experimental group, and the model group and the blank control group were gavaged with normal saline. The pathological changes of liver, spleen and bone marrow were observed by HE staining; Western blot and immunofluorescence (IF) were used to detect the expression of mitochondrial mitosis-associated proteins Drp-1, Fis-1 and mitochondrial autophagy-related proteins PINK1 and Parkin. Results: In the model group, leukemia cells infiltration was observed in the bone marrow, spleen tissue structure was destroyed, and focal degeneration and necrosis were seen in the liver. After the treatment of Shengma Biejia Decoction, the pathological damage in bone marrow, spleen and liver of mice was alleviated.Compared with the model group, the expression of mitochondrion division related protein Drp-1, Fis-1, autophagy related protein Pink1 and Parkin in bone marrow of mice treated with Shengma Biejia Decoction was significantly higher. Summary/Conclusion: Shengma Biejia Decoction has the effect of interfering with acute leukemia, and its mechanism may be related to increasing the expression of mitochondrial mitosis-related proteins Drp-1 and Fis-1, and the expression of mitochondrial mitosis-related proteins PINK1 and Parkin, thus promoting cell autophagy.
