摘要
The clenbuterole was extracted from samples by employing ethyl acetate and hydrochloric acid solution, and the cleaning procedure was performed by SCX solid phase extraction cartridge. The extract was warmed with water at 50 ℃ and blowed with nitrogen until it got near to dryness. Then it was derivated.Retention time and specificions were used to identify melamine. The selective fragments were m/z 86,212,262,277 respectively. The calibration curve of clenbuterol was linear(correlation coefficient: 0.999) within the range of 0.01~2.0 mg/L.The average recoveries at 0.01,0.10 and 0.20 mg/kg were 88.31%,92.85%,93.97%.The relative standard deviations(RSD) of the method was lower than 8% for each case, and the limit of detection(LOD),defined in terms of 3 times baseline nosie was 0.1 ug/kg respectively. The developing method was applied to study the metabolize behavior and mechanism of clenbuterol in pig.The results indicated that the residual of melamine was found in pig while starting to feed, the content of clenbuterol in pig was increased gradually, and the highest content of clenbuterol was 493.59 ug/kg,but the content of clenbuterol in pig was reduced gradually immediately while stoping feed.
