•First report of R-(−)-pranoprofen and S-(+)-pranoprofen differences in pharmacokinetics.•Pranoprofen enantiomers were quantified in rat plasma by a new LC-MS/MS method with per-4-chlorophenylcarbamate-β-cyclodextrin bonded chiral stationary phase.•The LLOQ for both pranoprofen enantiomers were 1.0 ng mL−1.•The chromatographic elution order of pranoprofen enantiomers were determined.
