•A photoelectrochemical biosensor based on highly signal-enhancement of MoOx/BiVO4-Ov is developed for DNA.•Coupling MoOx co-catalyst and oxygen vacancy on the BiVO4 can amplify signal.•The PEC biosensor exhibit excellent selectivity and sensitivity for DNA.
