Abstract
本文建立了基于超高效液相色谱-串联质谱(UPLC-MS/MS)的定坤丹中鹿茸的专属性鉴别及驯鹿茸投料的筛查方法.采用胰蛋白酶对定坤丹样品进行酶解处理,使用Waters Acquity UPLC BEH C18色谱柱(2.1 mm×100 mm×1.7μm)分离,以0.1%甲酸乙腈溶液和0.1%甲酸溶液为流动相进行梯度洗脱,流速0.3 mL/min,进样体积5 μL.电喷雾离子源(ESI)正离子模式,多反应监测(MRM)模式.结果表明,鹿茸专属性鉴别及驯鹿茸的各监测离子对专属性良好,30批实际样品均检出鹿源性成分;其中,8批检出鹿茸,其余22批均检出驯鹿茸,表明目前市场上定坤丹样品中鹿茸多来源于驯鹿.该方法的专属性好、操作简便快捷,进一步完善了定坤丹质量标准,为含鹿茸中药制剂中鹿茸的鉴别及驯鹿茸筛查提供参考.
