无菌水长期保藏黑木耳菌种性状研究

Abstract
以1994年利用无菌水保藏的44株黑木耳菌种和同时期同种4 ℃继代保藏黑木耳菌种为试材,采用生长状态测定、ISSR(inter-simple sequence repeat)、酯酶同工酶的方法,研究无菌水保藏方法对黑木耳菌种性状的影响,以期对黑木耳菌种的无菌水保藏方法优劣的判断提供参考依据.结果表明:44株黑木耳在无菌水中常温保藏28年,有36株保持活性,复活率达到81.8%,菌种继代培养后菌丝恢复正常生长状态,并能够正常出耳;与4 ℃保藏菌种相比,同一菌株间酯酶同工酶和ISSR,总体具有一致性,个别菌株具有一定的差异,不同菌株反映不同.
