Esomeprazole Floating Tablets by Using HUSK/HPMC Polymers: Formulation Development and In-vitro Characterization

The goal of the study was to develop and assess floating tablets with gastro-retentive properties by employing varying amounts of polymers such hydroxypropyl methyl cellulose (HPMC) and Husk. HPMC and Husk are capable of delaying the drug's release, allowing it to be delivered to a specific place in a regulated manner. Esomeprazole was selected as model drug owing to it relatively shorter half-life and bioavailabili-ty problems. By using the direct compression approach and sodium bicarbonate as a gas producing agent, various formulations were developed. Prepared formulations were evaluated for Pre compression (angle of repose, tapped density, bulk density, hausner's ratio and carr's index) and post compression tests (hardness ,thickness, friability, weight variation and content uniformity) In vitro testing of prepared formulations was conducted on the base of total floating time, lag time, percent swelling ,dissolution and determination of mechanism involved in drug release. Interestingly, the research exposed that the manufactured tablets from all formulations floated for even more than 12 h with controlled release patterns, demonstrating the viability of the examined approach. The optimum formulation F2, on the other hand, had the least floating time lag of 30 s and the longest drug release time of up to 12 h. Results of in vitro release confirmed that substantially enhancing the content of both polymers prolonged the release as well as buoyancy duration. Hence results showed that use of HPMC and HUSK can be good approach for development of esomeprazole sustained re-lease floating tablets.
