Abstract
目的:探讨HIF-1α、MT、Slug和E-cadherin在人乳腺癌中的表达及其意义.方法:采用免疫组化SP法,检测术前未使用放疗、化疗及激素治疗的69例乳腺癌组织中HIF-1α、MT、Slug和E-cadherin的表达情况.采用RT-PCR方法检测乳腺癌细胞系MCF-7和MDA-MB-231中HIF-1α、MT、Slug和E-cadherin mRNA的表达情况.结果:免疫组化结果表明:在69例乳腺癌组织中,HIF-1α、MT、Slug和E-cadherin的阳性率分别为71.01%、42.03%、43.48%和50.72%.HIF-1α、Slug和E-cadherin的表达对于TNM分期和腋淋巴结转移有显著性差异(P<0.05);MT的表达对于TNM分期有显著性差异(P<0.05).在69例乳腺癌组织标本中HIF-1α与MT表达呈显著正相关(r=0.285,P<0.05),MT与Slug呈显著正相关(r=0.379,P<0.01),Slug与E-cadherin呈显著负相关(r=-0.422,P<0.01).RT-PCR半定量结果显示:HIF-1α、MT、Slug和E-cadherin mRNA在人乳腺癌细胞系MCF-7和MDA-MB-231中均有表达,但在恶性程度更高的MDA-MB-231细胞中HIF-1α、MT、Slug表达较高,而E-cadherin表达较低,差异具有显著性(P<0.05).结论:HIF-1α、MT、Slug和E-cadherin在乳腺癌中的表达情况可作为监测乳腺癌发展的指标.
