•Autophagy is suppressed in CDDP resistant OS cells.•ANXA2 and Rac1 are upregulated in CDDP resistant OS cells.•Silencing ANXA2 or Rac1 re-activates autophagy and increases CDDP sensitivity.•ANXA2 and Rac1 modulate the osteogenic differentiation of OS cells.
