电针额区对血管性大鼠血清NSE、S-100β表达及海马回CA1区细胞凋亡的影响

Abstract
目的:观察电针额区对血管性痴呆(VD)模型大鼠血清神经元特异性烯醇化酶(neuron-specific enolase,NSE)、S-100蛋白钙结合蛋白β(S100 calcium binding proteinβ,S-100β)表达及海马回CA1区细胞凋亡的影响.方法:选取60只清洁级健康雄性Wistar大鼠,随机分为假手术组、模型组、西药组和电针组,每组15只.采用2-血管阻断法(2-VO)制作动物模型后,电针组先按照于致顺老师头穴分区法丛刺大鼠额区,再用脉冲电疗仪连接前庭和囟会穴,给予疏密波,20 min后出针,1次/d.西药组大鼠给予脑复康水溶液灌胃,1次/d.模型组与假手术组均灌胃给予生理盐水.然后将4组大鼠进行Morris水迷宫实验,观察治疗30 d后各组大鼠逃避潜伏期及穿越平台次数,并采用酶联免疫吸附法检测大鼠血清NSE、S-100β含量的表达.利用TUNEL法,观察光镜下海马回CA1区细胞凋亡状态.结果:与模型组比较,电针组与西药组逃避潜伏期都明显缩短(P<0.05),穿越平台次数都明显增高(P<0.05),血清NSE及S-100β含量都明显减少(P<0.05),细胞凋亡率都明显降低(P<0.01);电针组逃避潜伏期及穿越平台次数、血清NSE及S-l00β含量、细胞凋亡率与西药组比较,差异均无统计学意义(P>0.05).结论:电针额区能降低VD模型大鼠血清NSE、S-100β含量,减少海马回CA1区细胞凋亡数量,从而提高其学习记忆能力.
