产嗜热酯酶菌株的筛选、鉴定及发酵特性研究

Abstract
为获得可用于嗜热酯酶制备的高活性产酯酶微生物,满足果蔬制品精深加工用酶需求,本试验利用含三丁酸甘油酯的筛选培养基在堆肥高温期样品中筛选到一株高活性产嗜热酯酶菌株GRJ 2,对其进行了形态学、生理生化和分子生物学鉴定,并研究了其最适发酵条件.结果发现,经鉴定筛选到的GRJ 2菌株为嗜热淀粉芽孢杆菌(Caldibacillus thermoamylovorans).其最适生长温度为55℃;最适生长pH值为7,pH耐受范围5~9.菌种最佳活化时间为10 h,分批培养条件下嗜热酯酶最佳收获时间为16~22 h,及时补加新鲜培养液,可将产酶期延长4 h.菌株GRJ 2具有较高的产酯酶活性,酸碱耐受性强,可以用于工业酯酶的制备.
