•The combination of cell experiments and animal studies in this study, with the simultaneous application of cell lines and clinical specimens.•TCAF2 acts as an oncogene, associates with tumor immunity, and promotes the pathogenesis of glioma.•TCAF2 expression level is correlated with tumor grade and clinical outcome, and may serve as a therapeutic target and prognostic marker in glioma.
