•YIGSR and RGD peptides were conjugated to form a novel YIGSRGD peptide for MSC-based therapy.•Novel biotin-DFYIGSRGD hydrogel provides stability and structural support to MSCs.•Biotin-DFYIGSRGD enhances the anti-apoptotic and anti-inflammatory effects of MSCs.•Biotin-DFYIGSRGD improves the survival and therapeutic effects of MSCs.•The proposed hydrogel promotes kidney regeneration in AKI model.
