Abstract
目的 优化PARP1抑制剂AZD5305的合成工艺.方法 以6-甲基-5-硝基烟酸乙酯为原料,经过缩合、氧化、Wittig反应、氢化、脱氢、还原反应制备中间体3-乙基-7-(羟甲基)-1,5-萘啶-2(1H)-酮(8);以5-溴吡啶-2-甲酸甲酯为原料,经钯催化偶联、氨解、脱保护基制备中间体N-甲基-5-哌嗪-1-基-吡啶-2-甲酰胺(12);将中间体8氯代后与中间体12反应制得AZD5305.结果 与结论目标化合物结构经MS和NMR确证,优化后的工艺条件更加温和,操作简单,生产成本低,产品质量稳定可控.
