•Size of STING ligand nanocomplexes (NCs) is pH dependent, being 6-fold bigger than that of lysosomes at low pH.•Macrophage polarization from M2 to M1 upon STING ligand delivery might improve immune infiltration of cold tumors.•Biocompatibility and toxicity was assessed by hen egg CAM model.•Performed pilot in vivo STING ligand intratumoral administration studies on established B16.F10 tumor in mice.
