Detection of small molecules by extending the terminal protection to the polymerase

Biotin bound to SA adjacent to the 3′-terminus of the single-stranded DNA (ssDNA) produced a significant steric hindrance to TdT-catalyzed elongation. With the competition of free biotin, the SA could be released from the ssDNA and reveal the unprotected 3′–OH end for TdT elongation, which could achieve exponential fluorescence signal amplification.
