•Transient receptor potential vanilloid 1(TRPV1) is found in supramammillary nucleus.•Trpv1 mRNA is co-expressed with vesicular glutamate transporter 2 mRNA.•TRPV1 deficiency leads to less anxiety and depression-like behavior in mice.•TRPV1 antagonist induces anti-depressant effects in mice.•TRPV1 deficiency does not affect memory and learning capabilities of mice.
