395. Within- And Between-Cohort Test-Retest Reliability of Risky Decision-Making in the Iowa Gambling Task by the DAT Knockdown Mouse Model of Bipolar Disorder

The Iowa Gambling Task (IGT) is a cross-species translatable test of risky decision-making under uncertainty in which subjects choose between high risk/high reward (“risky”) and low risk/low reward (“safe”) options. People with bipolar disorder (BD) choose high risk/high reward options more frequently than healthy controls, consistent with risky decision-making behavior observed in this population. Cannabis may remediate risky decision-making in BD patients, but determining mechanisms, directionality, and effects of individual constituent compounds (e.g., cannabidiol; CBD) in humans remains difficult. The dopamine transporter (DAT) knockdown (KD) mouse model of BD mania recreates patients’ risky IGT behavior, thus enabling investigation of cannabinoid effects on BD-like decision-making. Establishing test-retest reliability of IGT performance and consistency of DAT KD behavior would facilitate translation of these data to the clinic.
