•A unique approach for detecting Ac-DL-Leu chiral separation in human fingernails.•Ac-DL-Leu has been efficiently quantified in the nails of HVs, PDs, and DPs.•Ac-DL-Leu levels in DPs’ nails were found to be much lower than in PDs and HVs.•This method provides a new strategy for diabetes detection.
