Abstract
目的 探讨加味温胆汤对抑郁模型大鼠肠道细菌丰度及构成的影响.方法 采用孤养结合慢性不可预见性温和应激(CUMS)法复制抑郁模型大鼠,通过应用旷场实验、糖水消耗实验,检测各组大鼠情绪变化,评估抑郁模型是否成功复制及加味温胆汤抗抑郁效应;应用16s rDNA高通量测序分析技术,研究加味温胆汤对肠道细菌丰度及构成的调控作用.结果 与空白组比较,抑郁模型大鼠旷场实验、糖水消耗量得分均减少(P<0.01),肠道有益菌Firmicutes和Bacilli丰度降低、有害菌Bacteroidetes和Bacteroidia丰度增加(P<0.05).与模型组相比,加味温胆汤干预后,大鼠旷场实验、糖水消耗量得分升高(P<0.05),肠道有益菌Firmicutes和Bacilli丰度增加、有害菌Bacteroidetes和Bacteroidia丰度降低(P<0.01),且相似度上更接近于空白组.结论 加味温胆汤可能通过增加益生菌抑制致病菌调控肠道菌群,从而发挥抗抑郁作用.
