Sugarcane Leaf Polysaccharide (Slp) Exerts a Therapeutic Effect on Cardiovascular Diseases Via Necroptosis

Background: Necroptosis, a type of programmed cell death with necrotic morphology featured by swelling of the cells, rupture of the plasma membrane, and dysfunction of the organelle, has been always observed in cardiovascular diseases. Sugarcane Leaf Polysaccharide (SLP) is a class of primary components of sugarcane leaves, which plays a favorable role in cardiovascular protection. However, the positive effect and exact mechanism of SLP remains to be illustrated on myocardial ischemia reperfusion (MI/R). In this study, the protective effects of SLP in vitro and in vivo on MI/R injury were investigated.Methods: TBHP-stimulated-H9c2 cell was used to investigate the protective effect of SLP on myocardial cells in vitro. While SD rat was used to investigate the protective effect of SLP on MI/R injury.Results: SLP significantly reversed TBHP-induced H9c2 cell death via inhibiting necroptosis and oxidative stress in vitro. The Nrf2/HO-1 pathway participated in SLP’s antioxidative activity. While SLP suppressed necroptosis by decreasing phosphorylation of RIP1, RIP3, and MLKL in TBHP-stimulated H9c2 cells. In vivo, SLP attenuated MI/R injury by decreasing myocardial infarct area, increasing MPO and SOD level, and decreasing MDA, IL-6, and TNF-α levels.
