•A periplasmic protein Cyt c4 is targeted to play a role in photoelectron transfer.•An ITO-bottom electrochemical reactor records the photoelectron uptake process in situ.•UV–Vis spectroscopy detects 3.35 times higher Cyt c at light than in the dark.•The abundance of Cyt c4 significantly increases with light intensity enhancement.
