PDF file - 407K, TMEM16A overexpression promotes cell proliferation in HEK-293T cells. TMEM16A overexpression induced cellular proliferation (*P<0.05). A representative immunoblot is shown (A). TMEM16A confers anchorage-independent viability in keratinocytes. Spontaneously immortalized oral keratinocytes were transduced with TMEM16A expressing virus or control (VC) and anchorage independent viability was evaluated. TMEM16A overexpression increases viability in anchorage independent conditions (B, p<0.05). TMEM16A knockdown induced growth retardation in SCCHN cells Cal33 and PCI-15B (C). Spontaneously immortalized wild-type MEFs expressing H-rasQ61L were transduced with retroviruses encoding TMEM16A or a hypo-morphic version of TMEM16A (TMEM16A-K610A). Focus formation was determined by crystal violet staining. TMEM16A overexpression induces a 3-fold increase in foci formation, the experiment was repeated several times in triplicate (D, p<0.05). Clonogenic assay was used to demonstrate increased colony formation after TMEM16A overexpression in T24, UM-SCC1 and immortalized keratinocytes (E).
