Abstract
以循环肿瘤细胞(CTC)的特异性膜蛋白EpCAM为识别受体,以羧基荧光素(FAM)标记的EpCAM核酸适体(Apt-FAM)为识别配体,当Apt-FAM特异性识别EpCAM后,细胞表面分布着大量的FAM荧光团,FAM荧光团的疏水性诱导非离子型表面活性剂Tween-80的疏水端在FAM周围聚积,形成以FAM为中心的伞形纳米胶束,从而增强了细胞膜的特异性识别荧光,提高了荧光信号的信噪比;利用数字信号放大技术,将成像细胞的膜荧光信号放大,使成像细胞的轮廓完整、清晰地呈现出来,实现了单细胞特异性识别轮廓的可视化,提高了捕获CTC计数和尺度估计的准确度.
