Abstract
目的:探讨circCPA4靶向miR-642a-5p对胃癌AGS细胞增殖、迁移和侵袭的影响.方法:RT-qPCR检测胃癌组织中circCPA4和miR-642a-5p表达水平.在AGS细胞中分别转染si-circC-PA4、si-NC、pcDNA、pcDNA-circCPA4、miR-NC、miR-642a-5p模拟物、si-circCPA4+anti-miR-NC、si-circCPA4+anti-miR-642a-5p.通过划痕愈合、集落形成、CCK-8、Transwell实验分析AGS细胞划痕愈合率、克隆数、活力、侵袭数.circCPA4和miR-642a-5p的相互作用通过双荧光素酶法确定.结果:circCPA4在胃癌组织中表达上调(P<0.05),miR-642a-5p表达下调(P<0.05).下调circCPA4表达后细胞划痕愈合率、克隆数、吸光度(A)值、侵袭数降低(P<0.05),miR-642a-5p表达水平升高(P<0.05).上调circCPA4表达后细胞miR-642a-5p表达水平降低(P<0.05).上调miR-642a-5p表达后细胞划痕愈合率、克隆数、A值、侵袭数显降低(P<0.05).circCPA4与miR-642a-5p直接特异性结合.干扰miR-642a-5p表达减弱下调circCPA4表达对细胞划痕愈合率、克隆数、A值、侵袭数的影响(P<0.05).结论:下调circCPA4通过促进miR-642a-5p表达来抑制胃癌AGS细胞增殖、迁移和侵袭.
