•Activated platelets secrete hemostatic proteins from alpha-granules.•We investigated how von Willebrand factor (VWF) and VWF propeptide (VWFpp) are released from platelet alpha-granules.•VWF and VWFpp are localized in the same eccentric alpha-granule subdomain in resting platelets.•VWF and VWFpp are differentially secreted from individual alpha-granules upon activation.
