•The first study on enantioselectivity of chlorphenesin in vitro and in vivo.•Significant enantioselective difference of chlorphenesin was observed in rat.•The cytotoxicity of S-chlorphenesin was apparently higher than that of R-chlorphenesin on HepG2 cells.
