•Design and synthesis of eighteen 2-phenylthiazole derivatives.•The new compounds were evaluated for cholinesterases inhibitory activity.•Compound 5a revealed the highest acetylcholinesterase inhibitory activity.•Molecular docking and molecular dynamics simulations study of compound 5a indicated that it interacted with PAS of AChE.
