Nucleation kinetics-based solvent selection for the liquid antisolvent crystallization of a lipophilic intermediate

摘要
•Impact of reactor geometry and stirrer type on the metastable zone width (MSZW) of lipophilic intermediate was verified.•Crystalline™ was found a reproducible platform for MSZW determination using Intermediate A in isopropanol as a model system.•A combined antisolvent addition and polythermal approach was applied to determine MSZWs.•The solvent selection order for LASC process of intermediate A, as per ease of nucleation: DMAC > NMP ≫ acetone > IPA.•Mean particle size determined from LASC process at similar driving forces confirm the rank ordering of solvents.
