The characteristics of SKOV-3/Luc cells. The human ovarian cancer cell line SKOV-3/Luc is known to be HER2-positive. A, Western blot analysis showed the expression of wt-HER2 protein (185 kDa) in SKOV-3/Luc cells, the same as in N87 cells. B, In flow cytometric analysis, the high HER2 expression level was confirmed in SKOV-3/Luc cells, as compared with in the parental cells. C, Immunohistochemistry showed diffuse HER2 expressions on SKOV-3/Luc cells. D, SKOV-3/ Luc cells seeded on 96-well plates at a density of 2ï‚'103 cells/ well for 24 h. Trastuzumab and Tra-IR700 were added (10 μg /mL) for 6 h, and NIR light was irradiated at 5 J/cm2. Forty-eight h after the irradiation, the cell viability was assessed. Tra-IR700-mediated PIT efficiently decreased the cell viability. *BF, bright field; ** P< 0.05 vs the other control group using a log-rank test.
