Abstract
运用RT-PCR技术获得罗氏沼虾(Macrobrachium rosenbergii)GIH-like基因的开放阅读框(ORF)序列,通过荧光定量PCR技术检测该基因在罗氏沼虾不同组织中的表达量,并运用原核表达技术诱导表达罗氏沼虾GIH-like成熟肽的重组蛋白.结果显示:罗氏沼虾GIH-like的ORF全长为339 bp,共编码112个氨基酸残基,其中包含34个氨基酸组成的信号肽及78个氨基酸组成的成熟肽,具有CHH家族Ⅱ型肽的典型特征;同源性和进化关系分析表明,罗氏沼虾GIH-like与日本沼虾GIH的同源性最高,亲缘关系最近;荧光定量PCR结果显示,罗氏沼虾GIH-like在眼柄中的表达量最高,极显著(P<0.01)高于其他组织的,在脑、神经节、心脏等组织中表达量较低;诱导表达的罗氏沼虾GIH-like重组蛋白的相对分子质量约为2.6×104,除去标签蛋白后约为8.5×103,与Prot Param预测的9.4×103接近.
