Abstract
•The chiral 1-hydroxy-cyclopentenyl analogs (22a-b) were synthesized in the multi-gram scale from D-ribose.•New N7-/N9-2,6-dihalo-3-deazaneplanocin and N7-6-azido-3-deazaneplanocin analogs were prepared.•Novel N9-2,6-dibromo-3-deazaneplanocin (44a) exhibited the most potent activity against flu a (H1N1) with EC50 value of 7.0 μM in vitro.
