Progress of the genetic improvement of the cowpea bean (Vigna unguiculata (L.) Walp) in the Colombian Caribbean (vol 27, 2022)

The cashew (Anacardium occidentale L.) in Colombia is concentrated on the Atlantic coast and the highlands of the Eastern Plains. One of the main problems that cashew production presents on the Atlantic coast is obtaining quality planting material, where plantations established with sexual seed of landrace materials currently predominant. In this respect, Agrosavia carried out the molecular identification of cashew trees in the municipality of Polonuevo (Atlantico) in order to enable this locality as a basic orchard for source of Mapiria Ao1 clone buds. For this, samples of plant tissue were taken from 72 selected trees based on phenotypic characteristics consistent with those of the Mapiria Ao1 clone. DNA extraction was performed following the protocol developed by Perez-Pazos and Berdugo with modifications. The DNA obtained was amplified by specific markers for the identification of cashew clones. The results of the analyzes with the seven combinations of SSR primers allowed the identification of 64 trees corresponding to clone Mapiria Ao1, additionally seven (7) trees of a genotype called "indeterminate Caribia" and 1 tree of clone 83-15 were identified. The results obtained allow us to be certain of the genetic identity of the trees analyzed, and would enable their use as a source of buds of the Mapiria Ao1 clone for the production and commercialization of certified plant propagation material.
