•Higher organic load and temperature increased CH4 production and substrate conversion.•Stoichiometric, kinetic, and thermodynamic biochemical routes were analyzed.•The best condition had a microbial structure more typical of a single-stage system.•Scale-up estimate single-stage system: 80 m³ and 9.89 MJ.kg-COD−1.•Scale-up estimate two-stage system: 123 and 12 m³ and 5.14 MJ.kg-COD−1.
