•The feasibility and advantages of the GUESS method for separation of complex samples from natural products was confirmed.•A methodology was developed to adjust the polarity of the CCC solvent system for the separation of several complex fractions obtained from Salvia bowleyana Dunn.•12 diterpenoids identified by one-step elution-extrusion CCC.
