•The effect of pH shift on the anti-CD52 mAb production process, quality and biopotency is investigated.•The robust production process was established by pH shift.•The quality of the mAb was improved by pH shift.•The biopotency of the mAb in vitro was also strengthened by pH shift.•The contents of galactosylation and afucosylation were positively related to CDC and ADCC efficacy.
