Abstract
采用实时细胞电子分析技术(real-time cell-based assay,RTCA),以大鼠心肌细胞(H9c2)建立复方丹参滴丸细胞生物电传感模型,测定其体外溶出与吸收的时间剂量依赖性细胞反应曲线(time/dose depended cell response profiles,TCRPs),以此建立连续动态溶出与吸收动力学模型.得到复方丹参滴丸的细胞指数(CI)溶出与吸收动力学曲线和动力学模型,溶出动力学最佳模型均为Weibull,溶出曲线相似因子f2大于50,吸收曲线相关系数大于0.95.同时以小鼠药效实验得到复方丹参滴丸小鼠尾出血药效时间百分率曲线,溶出、吸收及药效曲线均具有相关性(r>0.9).结果表明RTCA技术用于中药复方固体制剂溶出与吸收动力学研究,突破了唯成分论的局限,用于评价中药复方制剂质量及批间一致性具有创新性和可行性,为中药复方制剂质量控制提供新的思路与方法.
