•The characterization of adeno-associated virus (AAV)-based gene therapy products represents significant analytical challenges.•Recent advances in mass spectrometry have provided a means to characterize multiple attributes of AAV gene therapy products.•Current methodologies and emerging trends in MS analysis of AAV gene therapy products are presented in this review, including intact AAV vector analysis, capsid viral protein characterization and impurity analysis.
