•First demonstration of module assembly of cellular DNA hydrogel by click chemistry.•Biomimetic DNA-based cellular hydrogel exhibited cell growth and migration.•DNA microstructures effectively connect with multiple cells without considering cell type.•Depending on the concentration of histone treatment, the cell release rate of the cellular DNA hydrogel can be controlled.•Cellular DNA hydrogel to cover the irregular wound shape, it showed a successful cell therapy effect on the skin.
