Abstract
目的 探讨circATRNL1靶向miR-181a-5p对高糖诱导的肾小管上皮细胞(HK-2)损伤的影响.方法 将HK-2细胞分为对照(Con)组(5.5 mmol/L葡萄糖)、高糖(HG)组(25 mmol/L葡萄糖)、HG+pcDNA组(转染pcDNA,25 mmol/L葡萄糖)、HG+pcDNA-circATRNL1组(转染pcDNA-circATRNL1,25 mmol/L葡萄糖)、HG+anti-miR-NC组(转染anti-miR-NC,25 mmol/L葡萄糖)、HG+anti-miR-181a-5p组(转染anti-miR-181a-5p,25 mmol/L葡萄糖)、HG+pcDNA-circATRNL1+miR-NC组(转染pcDNA-circATRNL1和miR-NC,25 mmol/L葡萄糖)、HG+pcDNA-circATRNL1+miR-181a-5p组(转染pcDNA-circATRNL1和miR-181a-5p模拟物,25 mmol/L葡萄糖).RT-qPCR检测circATRNL1和miR-181a-5p表达;流式细胞术检测细胞凋亡率;ELISA试剂盒检测细胞培养上清中IL-6和TNF-α水平.双荧光素酶报告实验分析circATRNL1和miR-181a-5p的相互作用.结果 与Con组比较,HG组HK-2细胞凋亡率、miR-181a-5p相对水平、细胞培养上清中IL-6和TNF-α水平显著升高(P<0.05),circATRNL1相对水平显著降低(P<0.05).与HG+pcDNA组比较,HG+pcDNA-circATRNL1组HK-2细胞凋亡率、细胞培养上清中IL-6和TNF-α水平显著降低(P<0.05).与HG+anti-miR-NC组比较,HG+anti-miR-181a-5p组HK-2细胞凋亡率、细胞培养上清中IL-6和TNF-α水平显著降低(P<0.05).circATRNL1与miR-181a-5p直接特异性结合.与HG+pcDNA-circATRNL1+miR-NC组比较,HG+pcDNA-circATRNL1+miR-181a-5p组HK-2细胞凋亡率、细胞培养上清中IL-6和TNF-α水平显著升高(P<0.05).结论 circATRNL1靶向miR-181a-5p可抑制高糖诱导的肾小管上皮细胞凋亡和炎症损伤.
